摘要 目的:确定甲紫对白念珠菌毒力因子分泌型酸性蛋白酶和细胞外磷脂酶活性及芽管生成率的作用。方法:采用蛋黄平皿培养基和小牛血清白蛋白培养基分别检测筛选蛋白酶、磷脂酶阳性且Pz值≥2+的白念珠菌各12株,将2μg/mL、0.5μg/mL的甲紫溶液作用于各菌株菌悬液1h。分别测定不同浓度甲紫溶液作用前后各菌株蛋白酶、磷脂酶的Pz值及芽管生成率,比较甲紫溶液作用前后各菌株蛋白酶、磷脂酶活性及芽管生成率的不同。结果:2μg/mL、0.5μg/mL药物作用组磷脂酶活性、蛋白酶活性及芽管生成率均显著低于药物作用前对照组(P<0.05)。结论:低浓度甲紫溶液短时间作用于白念珠菌可降低菌株磷脂酶、蛋白酶活性和芽管生成率,从而降低白念珠菌毒力和致病性。 Objective: To study the effects of gentian violet (GV) on activity of secretory acid proteinase, extracellular phospholipase and germ tube formation rate of Candida albicans. Methods: Bovine serum albumin agar (BSA) and egg yolk agar (EYA) were used to detect the activities of proteinase and phospholipase of C. albicans respectively. Both 12 proteintase - positive and 12 phospholipase - positive isolates with 2 + score of Pz value or more were obtained. After a brief exposure (60 min) to sub - therapeutic concentrations (0.5μg/mL and 2 μg/mL) of GV the Pz value of proteinase and phospholipase and germ tube formation rate for two exposed groups and one unexposed control group were determined. Results: The activities of proteinase, phospholipase and germ tube formation rate in two groups exposed to GV were significantly lower than those in the group unexposed to GV ( P 〈 0.05). Conclusion: The results of this study indicate that sub - therapeutic concentrations of GV may decrease the candidal pmteinase, phospholipase activities and germ tube formation rate and then may suppress virulence and pathogenicity of C. albicans.
