摘要
构建了Anabaena PCC7120的复制起始蛋白Dna A的重组表达质粒,在Escherichiacoli中诱导其超量表达,纯化后免疫家兔获得抗体,采用western blotting检测Dna A在营养细胞和异形胞中的表达情况.结果显示,两种不同类型的细胞中都能够检测到Dna A的表达,而且在不同缺氮诱导时间,异形胞中Dna A的表达存在差异.说明DNA复制起始与异形胞的发育可能存在着一定的关系.
The corresponding antiserum against Dna A proteins of Anabaena PCC7120 were collected by injecting rabbits with purified proteins after over-expression from recombinant expressing plasmid in Escherichia coli. Then the antiserum was used to detect the expression of Dna A in vegetative cell and heterocyst by Western blotting. The result showed that Dna A could be detected in both type cells, and moreover, the expression of Dna A is different in different period heterocysts induced with nitrogen stepdown. It indicated certain relations exist between DNA replication and heterocyst differentiation.
