峨眉拟单性木兰多酚氧化酶活性与总酚含量的初步研究

摘要 以峨眉拟单性木兰嫩枝茎段为测试材料,研究多酚氧化酶(PPO)活性和总酚含量及其季节变化规律。结果表明,测定峨眉拟单性木兰PPO活性的最适pH值为7.0,最佳底物为0.14 mol.L-1的邻苯二酚,最佳温度为25℃;峨眉拟单性木兰PPO活性在生长期明显高于休眠期;峨眉拟单性木兰总酚含量在每年的4月、5月、9月3个时段最低,为无性繁殖取材最佳时间。 Taking burgeon and stem of Parakmeria omeiensis Cheng as test materials, polyphenol oxidase(PPO), total phenol content, and seasonal change law were studied. The results showed the suitable pH value for determining P. omeiensis PPO aetivit was 7.0, the optimal substrate was 0.14 mol.L^-1 of oxophenic acid, the optimal temperature was 25 ; P. omeiensis PPO enzyme activity at the growth stage was higher than dormancy stage, P. omeiensis total phenol content was lowest in Apil, May, and September in a year, and which was the optimal time for getting the cloning stuffs.
