嗅成鞘细胞提取液脑室内注射对大鼠阿尔茨海默病的治疗作用

Effects of distilled fluid of olfactory ensheathing cells by intracerebreventricular injection on treatment of Aizheimer' s disease rat

摘要 目的探讨嗅成鞘细胞(OECs)提取液脑室内注射对大鼠阿尔茨海默病的治疗作用。方法SD大鼠双侧海马注射Aβ1-40建立AD大鼠模型。实验动物分为4组:正常对照组、AD模型组、人工脑脊液注射组、OECs提取液注射组,每组8只。运用行为学测试、NADPH—d组化、刚果红组织学染色结合积分吸光度测定等技术,观察、比较各组AD模型大鼠注射4周后学习记忆能力、一氧化氮合酶(NOS)阳性神经元数以及老年斑的变化。结果①行为学测试:OECs提取液注射组大鼠达到学会标准所需训练次数(TN)、错误反应次数(EN)、潜伏期(D)、全天总反应时间(TRT)分别为(18.82±2.64)次、(4.81±1.85)次、(5.18±4.86)S、(1236.78±97.26)S,均明显少于和短于AD模型组[(48.33±5.10)次、(10.83±2.98)次、(11.32±12.23)S、(2264.32--244.60)S(P=0.000)]。②NADPH—d组化反应:OECs提取液注射组NOS阳性神经元数为(16.08-2.51)个/视野,比AD模型组(5.08±0.89)个/视野明显增多(P:0.000)。③刚果红染色及其吸光度(SP—IA)测定:OECs提取液注射组为(74.39±9.14),比AD模型组(481.15±144.18)明显减少(P=0.000)。④人工脑脊液注射组与AD模型组间上述指标均差异无显著性。结论嗅成鞘细胞提取液脑室内注射对大鼠阿尔茨海默病具有明显的治疗作用。 Objective To probe the effects of distilled fluid of olfactory ensheathing cells (DFOECs) by intracerebreventricular injection on treatment of Alzheimer' s disease(AD) rat. Methods The model of AD rats were builded by intraeerebreventrieular injection of Aβ1- 40.The rats were divided into the normal control groups, the model of AD ones,the manpower cerebral spinal fluid (MCSF) injected ones and the DFOECs injected ones at random, and there were eight rats every group. Four weeks after being injected, the change of memory recovery was observed by Y maze test,the total number of nitric oxide synthase positive neurons and senile plaques of hippocam- pus were observed by NADPH-d and Congo red histochemical staining. Results (1)Y maze test: TN,EN,D,TRT of DFOECs injected group obviously decreased compared to the model of AD group (P = 0. 000). The former was separately(18.82±2.64) times,(4.81 ± 1.85)times,(5. 18 ±4.86)s,(1236.78 ±97. 26 ) s, the latter was (48.33 ±5.10)times,(10.83 ±2.98)times,(ll.32±12.23)s,(2264.32-±244.60)s. (2) NADPH-d histo- chemical staining: the total number of NOS positive neurons of DFOECs injeeted group were higher than that of the model of AD group (P = 0. 000) , they were separately ( 16.08 ± 2.51 ) entries/eye shot, ( 5.08 ±O. 89 ) entries/ eye shot. (3) Congo red histochemieal staining:compared to the model of AD group and the DFOECs injected ones were signifieantly ehanged in SP-IA(P=0. 000) ,they were separately(481. 15 ± 144. 18) , (74.39 ±9. 14), (4) There was not signifieant differenees between the model of AD group and MCSF injeeted group. Conclusion The distilled fluid of olfaetory ensheathing eells can greatly improve the effeets on treatment of Alzheimer' s disease rat by intracerebreventricular injection.
