圆齿野鸦椿芽继代增殖的影响因素研究 被引量:12

摘要 [目的]研究不同激素配比、蔗糖、活性炭等附加物质及光照对圆齿野鸦椿丛生芽增殖的影响,寻求最适圆齿野鸦椿试管苗增殖的培养基配方及培养条件,为圆齿野鸦椿组培快繁提供一些有益的参考。[方法]以圆齿野鸦椿初代培养的丛生芽为外植体,基础培养基为1/2 MS,在不同激素配比、蔗糖、活性炭等附加物质的培养基中进行继代增殖培养,观察不同培养基及光照对圆齿野鸦椿试管苗芽增殖数量和玻璃化的影响。[结果]当培养基的激素配比为IBA 0.1 mg/L+6-BA 2.0 mg/L+NAA 0.1 mg/L+琼脂7 g/L+蔗糖30g/L,丛生芽增殖系数最高可达3.9倍,激素对芽增殖系数的影响为:NAA>6-BA>IBA;活性炭有抑制圆齿野鸦椿芽增殖、促进芽体高生长和降低玻璃化的作用;糖是增殖培养阶段不可缺少的成分;自然光照较日光灯照和暗培养效果好。[结论]激素的合理配比、蔗糖以及自然光照有利于圆齿野鸦椿芽继代增殖培养,活性炭有抑制圆齿野鸦椿芽继代增殖的作用。 [ Objective ] The research aimed to study the effects of hormone, cane sugar, active carbon and the others cultivated conditions on effective multiplication and illumination on clump bud generation of Euscaphid konishii. And find the most suitable formula of medium and culture conditions, and provide references for tissue rapid propagation of Euscaphid konishii. [ Method ] Taking initially cultured multiple shoot of Euscaphid konishii as the explants, successive transfer culture was conducted on 1/2 MS and culture medium including different hormone combination, cane sugar, active carbon and the other additional substances, and the influences of different culture mediums and illumination acted on multiplied quantity and glass transition of Euscaphid konishii were observed. [ Result ] The adventitious buds proliferation coefficients reached 3.9 times on 1/2 MS + IBA 0.1 mg/L + 6-BA 2.0 mg/L + NAA 0.1 mg/L + agar 7 g/L + sucrose 30 g/L, the hormones affected multiplication ratio was NAA 〉 6-BA 〉 IBA ; the active carbon could inhibit proliferation of Euscaphid konishii, promote height of brood body and decrease vitrification; cane sugar was the important components in multiplication culture stage; natural lighting was superior to fluorescent light and darkness culture. [ Conclusion ] Suitable hormone combination, cane sugar and natural lighting were useful to subculture multiplication culture, but the active carbon inhibited subculture multiplication of Euscaphid konishii.
