摘要
目的观察葛根异黄酮对甲基-苯基-吡啶离子诱导的帕金森病细胞模型P53基因表达的影响。方法将葛根异黄酮溶解在培养基使终浓度为50mg·L-1、100mg·L-1和200mg·L-1。并设不加药物的阴性对照组。PC12细胞与葛根异黄酮共孵育30min后,加入含250μmol·L-1MPP+的DMEM培养基,继续孵育96h。采用实时定量RT-PCR法检测P53基因表达。结果与对照组比较,MPP+组P53 mRNA表达上调(P<0.05),各浓度的葛根异黄酮组P53 mRNA表达下调(P<0.05)。结论葛根异黄酮对甲基-苯基-吡啶离子诱导的帕金森病细胞模型P53 mRNA的表达具有下调作用。
Objective To observe the effect of total isoflavones from pueraria lobata (TIP) on P53 mRNA expression in PD model cells induced by 1-methyl-4-phenylpyridinium ion (MPP +). Methods TIP was dissolved in culture medium at a final concentration of 50 mg·L -1 ,100 mg·L -1 and 200 mg·L -1 .Some cells (control) were exposed to culture medium.TIP was added to PC12 cells 30 min prior to the administration of MPP+. TIP and MPP + remained in the culture medium for 96 h.P53 mRNA expression were assayed by real-time quantitative reverse transcription-PCR. Results The P53 mRNA expression was up-regulate expression in MPP + groups compared with the control group (P<0.05). The P53 mRNA expression being down-regulated in TIP group compared with the MPP + group (P<0.05). Conclusions TIP down-regulated the P53 mRNA expression in PC12 cells induced by MPP +.
