摘要
[目的]弄清INHα前体基因与山羊繁殖季节性的关系,并研究INHα前体基因进化的保守性。[方法]对常年发情的山羊品种海门山羊和季节性发情的山羊品种安徽白山羊共20只母羊的抑制素0【前体基因(INHα)5’侧翼区和外显子进行了克隆和序列比较分析。[结果]与安徽白山羊相比,海门山羊INHα前体基因存在3个SNP,不存在氨基酸改变,核苷酸同源性为99.7%,氨基酸同源性为100%。在山羊、牛、猪、人、鸡、马、大鼠、狗之间INHα前体基因的核苷酸序列同源性为12.7%-96.5%。[结论]INHα前体基因在物种内趋于高度保守,任何核苷酸和氨基酸的改变都可能直接影响整个基因编码和基因调控能力的改变。
[ Objective ] This study was to reveal the relationship between Inhibin (INH)α precursor gene and seasonal character of goat reproduction and study the conservation of INHα precursor gene. [ Method] Twenty ewes of nonseasonal estrous breed (Haimen goat) and seasonal estrous breed( Anhui white goat) were used as experimental animals to clone the 5' flanking region and exon of INHα precursor gene, whose sequences were then analyzed. [ Result] Compared with Anhui white goat, there were three SNPs inα precursor gene of Haimen goat and no amino acid change. Haimen goat and Anhui white goat shared the homologues of 99.7% in nucleotide and 100% in amino acid sequence of INHα precursor gene. The homologues of nucleotide sequences of lNHα precursor gene among goat, cow, pig, human, chicken, hors'e, rat and dog were from 12.7% to 96.5%. [ Conclusion] INHα precursor genes tends to be highly conservative within a species, any change in nucleotide or amino acid could directly influence gene coding and its regulating capacity.
