上皮性卵巢癌中PTEN和HIF-1α的表达及其相关性 被引量:6

摘要 目的检测上皮性卵巢癌组织中PTEN和HIF-1α表达情况,揭示抑癌基因PTEN的丢失可以激活HIF-1α表达,并显示PTEN和HIF-1α的表达与上皮性卵巢癌的恶行程度和预后的关系,以及二者在上皮性卵巢癌中的相关性。方法应用SP免疫组织化学法检测37例上皮性卵巢癌、17例正常卵巢组织和19例卵巢交界性肿瘤。结果PTEN在正常卵巢组织和交界性卵巢肿瘤组织中有表达,在高分化卵巢癌中有少量表达,而在中低分化卵巢癌中表达明显减少或消失;HIF-1α在正常卵巢或卵巢上皮性交界瘤中无表达,而在高分化卵巢癌中有少量表达,中低分化卵巢癌组织中表达明显增多。PTEN和HIF-1α与卵巢的恶性程度和转移有明显关系。结论PTEN表达缺失而HIF-1α表达明显增强,在上皮性卵巢癌恶性进程中起重要作用,可以推测PTEN表达的缺失诱导了HIF-1α的表达,二者在恶性卵巢癌的肿瘤形成和预后判断中呈负相关。 Purpose To investigate the expression of PTEN and HIF-1α in epithelial ovarian carcinoma and the relationship of expression of PTEN or HIF-1α with epithelial ovarian carcinoma malignancy progression and prognosis, as well as the correlation between PTEN and HIF-1α in epithelial ovarian carcinoma. Methods HIF-1α and PTEN were detected in 37 cases of epithelial ovarian carcinoma, 17 normal ovarian tissue and 19 borderline malignancy by immunohistochemistry using streptavidin-peroxidase (SP) method. Resuits Expression of PTEN was observed in normal ovarian tissue and borderline malignancy tissues, but the expression decreased in well-differentiated ovarian carcinoma and absence of the expression in intermediate and poorly-differentiated tumors. No expression of HIF-1α was found in normal and borderline tumor tissue, the expression of HIF-1α decreased in well-differentiated, but in poorly-differentiated it expressed significantly increased. The expression of HIF-1α was significantly correlated with malignant progression and metastasis. Conclusions Loss of PTEN and the overexpression of HIF-1α may play an important role in the development of epithelial ovarian carcinoma and metastasis. It can be supposed that loss of PTEN is associated with induction of HIF-1α expression in epithelial ovarian carcinoma. The two factors are negatively correlation in tumor malignant progression and prognosis.
