连翘提取物对脓毒血症小鼠及体内T淋巴细胞影响 被引量:10

Effects of forsythia suspensa extract on the sepsis of mouse induced by cecum ligation and puncture and T lymphocytes in vivo

摘要 探讨连翘提取物(FS)对盲肠结扎穿孔(CLP)诱导脓毒血症小鼠的影响以及FS对CLP诱导小鼠脓毒血症体内T淋巴细胞的影响。建立脓毒血症动物模型,观察7 d内FS对小鼠存活率的影响;计算各组小鼠胸腺和脾脏指数;MTT比色法以及CFDA-SE染色分析,流式细胞术(FCM)检测FS对机体T淋巴细胞增殖的影响。FS可延长CLP诱导脓毒血症小鼠的存活率;FS药物治疗组小鼠脾脏的重量和指数明显增加和小鼠胸腺指数明显增加;药物FS治疗组对脓毒血症早期炎症性小鼠淋巴结细胞的增殖有抑制作用。FS对CLP小鼠胸腺和脾脏有保护作用。对脓毒血症早期炎症引起的T淋巴细胞增殖有抑制作用。 To investigate the effect of forsythia suspensa (FS) extract on the sepsis mouse induced by cecum ligation and puncture (CLP) operation and the effect on T lymphocytes of the sepsis model in vivo, the experimental model of sepsis was established by CLP operation, and the mortality rate of mouse sepsis model in 7 days and the thymus and spleen indexes of mouse were calculated. After the staining with CFDA-SE, the proliferation-related index of mouse lymphocytes was analyzed by flow cytometry as well as MTT. The result of the in vivo studies demonstrated that FS extract could protect mice against lethality after CLP operation. Compared with CLP treated mice, the weight of spleen and thymus indexes were significantly increased in the FS extract-treated mice. In addition, FS extract could show the inhibitory effect on the proliferation of T lymphocytes in early state of inflammation of sepsis. It is concluded that FS can protect mouse thymus and spleen by sepsis from destruction of and it can also inhibit the proliferation of T lymphocytes in early state of inflammation in vivo.
