大黄酸对转化生长因子β1诱导的人肾小管上皮细胞血小板反应蛋白1表达的影响 被引量:1

摘要 目的探讨大黄酸对转化生长因子β1(transforming growth factor β1,TGF-β1)诱导的人肾小管上皮细胞(HK-2)血小板反应蛋白1(thrombospondin-1,TSP1)表达的影响。方法体外培养细胞,观察TGF-β1(4ng/ml)诱导下低、中、高浓度的大黄酸(10μg/ml、20μg/ml和40μg/ml)对HK-2细胞TSP1 mRNA和蛋白表达的影响。Real Time PCR检测TSP1 mRNA表达,Western Blot检测TSPI蛋白表达。结果与空白对照组比较,TGF-β1(4ng/ml)明显上调HK-2细胞TSP1 mRNA和蛋白表达。与TGF-β1(4ng/ml)阳性对照组比较,中浓度和高浓度大黄酸明显抑制了TSP1 mRNA和蛋白表达。结论大黄酸能抑制TGF-β1诱导的HK-2TSP1 mRNA和蛋白表达。 Objective To investigate the effect of rhein on the expression of thrombospondin-1 in renal tubular cells induced by transforming growth factor β1. Method HK-2 cells were stimulated by different concentrations of rhein(10,20, and 40 μg/mi) in the presence of TGF-β1 (4 ng/ml), untreated HK-2 cells served as normal control group,and HK-2 cells stimulated by TGF-β1 (4 ng/ml) as positive group. Real Time PCR and Western Blot were used to detect the expression of TSPI mRNA and protein,respectively. Result The expression of TSP1 mRNA was up-regulated obviously in HK-2 cells when stimulated by TGF-β1 (4 ng/ml). The protein expression of TSP1 was also up-regulated simultaneously in HK-2 cells when stimulated by TGF-β1 (4 ng/ml). The expression of TSP1 mRNA and protein induced by transforming growth factor 61 was down-regulated markedly with 20 μg/ml and 40 μg/ml of rhein. Conclusion Rhein can inhibit the expression of TSP1 mRNA and protein.
