摘要
目的探讨补肾排毒合剂对肾间质纤维化的作用机制。方法将60只清洁级昆明小鼠随机分为正常组(I组)、UUO组(Ⅱ组)、补肾排毒合剂组(III组)和洛汀新组(IV组)。采用单侧输尿管结扎术建立肾间质纤维化模型。术后7、14、21、28d处死小鼠,取手术肾组织行HE染色,免疫组织化学方法测定肾组织中纤连蛋白及整合素α2、β1的变化。结果纤连蛋白及整合素α2、β1在正常肾组织中微弱表达,Ⅱ组表达较I组升高,Ⅲ组和Ⅳ组各时间点的表达较Ⅱ组降低,Ⅲ组和Ⅳ组之间无明显差异。结论补肾排毒合剂可通过调节纤连蛋白及整合素α2、β1的表达延缓肾间质纤维化的进展。
Objective To investigate the action mechanism of Bushen Mixture in renal interstitial fibrosis. Method Murine renal interstitial fibrosis models were induced by unilateral ureteral obstruction(UUO). Sixty kunming mice were randomly divided into normal control group(group I ), UUO group (group II ),Bushen Mixture-treated group(group III ) and lotensin-treated group (group IV ). After 7,14,21, and 2B days the mice were killed, and renal tissue from the operative side was subjected to HE staining. By using immunohistochemistry, fibronectin and integrin α2, β1 in renal tissue were detected. Result Fibronectin and integrin α2, β1 in normal kidney tissues had a weak expression. The expression levels of fibronectin and integrin α2, β1 in group 11 were higher than in group, and those in groups III and IV at each time point were lower than in group II,but there was no significant difference between groups III and IV. Conclusion Bushen Mixture can slow renal interstitial fibrosis through regulating the expression of fibronectin and integrin α2, β1.
