Optimization of Compatibility Ratio of Rhizoma Zingiberis and Pericarpium Citri Reticulatae Extracts for Counteracting Rats Myocardial Ischemia by Uniform Design

摘要 目的观察干姜、陈皮提取物对异丙肾上腺素(ISO)致大鼠急性心肌缺血的保护作用,并探讨其最佳剂量配比。方法运用均匀设计方法,以大剂量ISO建立大鼠急性心肌缺血模型,测定24h大鼠血清肌酸激酶(CK)的含量,以优化干姜、陈皮提取物抗心肌缺血的剂量配比。结果与对照组相比,模型组的CK水平显著升高(P<0.001);与模型组相比,各实验组均能在一定程度上降低CK水平。经多元回归分析,干姜、陈皮提取物最佳剂量组合为0.255mL/kg、0.141mL/kg。结论干姜、陈皮提取物配伍对大剂量ISO致大鼠心肌缺血具有保护作用。两者最佳配比约为2∶1。 Objective To observe the protective effects of the extracts of Rhizoma Zingiberis and Pericarpium Citri Reticulatae on myocardial ischemia reduced by isoproterenol (ISO) in rats, and to explore their optimal ratio. Methods Acute myocardial ischemia rat model was established by continuously subcutaneous injection of large - dose. Then the content of the serum creatine kinase (CK) within 24 hours in rats were detected, and the optiaml ratio of Rhizoma Zingiberis and Pericarpium Citri Reticulatae was optimized by uniform design. Results Compared with the control group, the content of CK in the model group was significantly increased ( P 〈 0. 001 ). And compared with the model group, the content of CK in experimental groups was lower than that in the model group ( P 〈 0.01 or P 〈 0. 001 ). The results of multiple regression analysis showed that the optimal ratio of the two extracts was 0. 255 mL/kg of Rhizoma Zingiberis and 0. 141 mL/kg of Pericarpium Citri Reticulatae. Conclusion The extracts of Rhizoma Zingiberis and Pericarpium Citri Reticulatae have obviously protective effect on the myocardial ischemia induced by isoproterenol, and the optimal ratio of the extracts from Rhizoma Zingiberis to the extracts of Pericarpium Citri Reticulatae is 2 : 1.
