摘要
目的研究3'-大豆苷元磺酸钠(DSS)对氧化低密度脂蛋白诱导脐静脉内皮细胞损伤的保护作用及其机制。方法体外培养脐静脉内皮细胞,将细胞分为空白对照组、氧化损伤对照组(ox-LDL)、氧化损伤加入维生素E对照组(VE+ox-LDL)、氧化损伤加入DSS低、中、高浓度组(DSS-L、DSS-M及DSS-H组)。将ox-LDL作用于预先加入VE及不同浓度DSS预处理24 h的内皮细胞,继续培养24h,MTT法检测细胞活力,比色法检测丙二醛(MDA)及一氧化氮(NO)的含量,检测乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果预先加入VE、中高浓度的DSS可提升损伤的内皮细胞的活力,可显著降低损伤内皮细胞的MDA及NO的释放,可显著提升损伤细胞的LDH、NOS、SOD和GSH-Px的活性,但低浓度的DSS对上述指标影响不大。结论DSS可减轻ox-LDL对血管皮细胞的氧化损伤,起到一定的抗氧化保护作用。
Objective To investigate the protective effect and mechanisms of 3′- daidzein sulfonate sodium on vascular endotheli- al cell injury induced by oxidized low - density lipoprotein. Methods Human umbilical vein endothelial cells (HUVECs) were cuhured in vitro and divided into 5 groups : control, ox - LDL, VE + ox - LDL, DSS - L, DSS - M and DSS - H groups. Endotheli- al cells were incubated with ox - LDL for 24 hours after they were incubated with Vitamin E(VE) and different doses of DSS for 24 hours. The proliferation of HUVECs was assayed by the 3 - (4,5 -dimethylthiazol -2 -yl) -2,5 -diphenyhetrazolium bro- mide method, and the levels of NO and MDA, the activities of LDH, SOD, NOS and GSH - PX were measured. Results The level of MDA and the activity of LDH in ox - LDL damaged group were remarkably higher than those in VE, DSS - M and DSS - H groups. The activities of SOD, NOS, GSH - PX and the concentration of NO in ox - LDL damaged group were lower than those in VE,DSS - M and DSS - H groups. There was no significant difference between DSS - L group and ox - LDL group. Conclusion 3′- daidzein sulfonate sodium has protective effect on HUVECs injury induced by ox - LDL, which may be related to increasing the antioxidant capacity of HUVECs.
