摘要
目的研究拳参正丁醇提取物对脑缺血再灌注损伤的保护作用,并初步探讨其保护机制。方法大鼠大脑中动脉栓塞,制作脑缺血再灌注损伤模型,缺血后10 min,于舌下静脉给予不同剂量的拳参正丁醇提取物。再灌注24 h后,断头取脑,TTC染色,测定脑梗塞体积;取血,离心后,取血清,比色法测血清NOS、SOD的活性及NO、MDA的含量。结果与模型组相比,拳参正丁醇提取物治疗组脑梗塞体积减小,血清NOS和SOD的活性升高,NO含量升高,MDA含量降低。结论拳参正丁醇提取物对脑缺血再灌注损伤具有一定的保护作用,其保护机制与拳参正丁醇提取物促进NO释放及增强氧自由基的清除有关。
Objective To explore the therapeutic effects of n - Butyl alcohol extract of Polygonum bistorla L. on cerebral ischemia/reperfusion injury and its mechanism. Methods A rat cerebral ischemia/reperfusion injury was made through embolization of the middle cerebral artery, and the injury was treated with n - Butyl alcohol extract of Polygonum bistorla L. , using sublingual vein injection. The cerebral tissue was stained using the TTC method, and the infarction volume was investigated. Moreover, the activities of NOS and SOD, and the levels of NO and MDA were also measured. Results After treatment, a reduction in the infarction volume and elevation in the activities of NOS and SOD in serum were observed. An elevation in NO level and a decrease in MDA level were also observed. Conclusion n - Butyl alcohol extract of Polygonum bistorla L. has certain therapeutic effects on cerebral ischemia/reperfusion injury by enhancing NO release and scavenging oxygen free radical.
