草杞颗粒对荷瘤小鼠肿瘤细胞凋亡的影响

Study on the Tumor Inhibitory Effect of Caoqi Granule

目的草杞颗粒对荷瘤小鼠的抗肿瘤作用机制。方法取昆明种小鼠18～22g，雌雄各半，共50只，接种s，80肿瘤细胞在腋窝皮下0．2ml，然后分组为：阴性组（空白组），24h后灌胃给水0．5ml／20g，1次／d，连续8d；阳性组，灌胃给5氟尿嘧啶（5-Fu）25mg／kg；草杞颗粒小剂量组生药2．21g生药／kg，中剂量组4．43g／kg，大剂量组8．85g生药／kg，上述每组均在接种肿瘤后24h灌胃给药，1次／d，连续8d，次日称重、剥离瘤块，入10％中性福尔马林固定。用免疫组化法观察促凋亡免疫酶标抗体Bax、Bcl-6、Hsp27的表达。结果草杞颗粒组均见有大片肿瘤细胞坏死、凋亡，小组累计坏死分数在小、中、大剂量组分别为2．28，2．50和2．76，坏死程度随剂量增大呈线性增加关系。促凋亡免疫酶标抗体Bax、Bcl－6、Hsp27较5－Fu组（1．74）有明显的阳性强度表达增加。结论草杞颗粒能促进肿瘤细胞凋亡，抑制S180肿瘤细胞生长周期。

Objectlve To observe the anti -tumor effect and explore the possible mechanism of Caoqi granule on tumor bearing mice. Methods The mice were transplanted by armpit injection of S180 tumor cells, and divided into positive control（5 -FU）, low dose group （Caoqi keli,2.21 g/kg） ,middle dose group （Caoqi keli,4.43 g/kg） and high dose group （Caoqi keli,8.85g/ kg）. Using immnunohistochemical staining technique, we detected the apoptotic related gene Bax, Bcl -6, Hsp27 protein expression. Results The phenomena of a large of tumor cell death and apoptosis were observed. The death index of three therapy groups with Caoqi keli was 2.26（low） ,2.50（middle） and 2.76（high） ,which was higher than that of the positive group（1.74）. The expression of apoptotic related gene Bax, Bcl - 6, Hsp27 were higher than that of model and positive group. Conclusion Caoqi granule can induce tumor cell death and apoptosis and inhibit the growth of S180 tumor cell line in vivo.
