龙胆药材提取工艺的正交实验设计法优选 被引量:7

摘要 目的建立龙胆最佳提取工艺,为龙胆的种植和质量研究奠定基础。方法采用L9(34)正交表安排实验,确定最佳提取条件。色谱柱为Shimadzu C18(3.0 mm×75 mm,1.7μm);流动相为乙腈-0.4%磷酸(V∶V=5.5∶94.5),流速0.80ml/min;检测波长235,270 nm;柱温为40℃。结果影响因素最大的是提取溶剂,其次是提取方法,最小的是提取时间。结论各因素较优的水平组合为A1B1C1,即甲醇超声提取30 min。 Objective To establish best extraction process of Gentiana scabra Bge. by UPLC so as to provide base for the plant and research of Gentiana scabra Bge. Methods By L9 ( 3^4 ) orthogonal design , the optimal condition was confirmed as follows : Shimadzu C18(3.0 mm ×75 mm,1.7μm), Acetonitrile -0.1% Phosphorylation(V: V = 5.5: 94.5) as nwbile phase the flow rate was 0.8 ml/min, detection wavelength was 235nm and 270nm and the columne temperature was 40℃. Results The extraction solution was the most effective factor , the second was the method, and the last was the extraction time. Conclusion The best extraction process is A1 B1C1 : super sonic extraction for 30 min in methanol.
