摘要
目的:观察中药肾康宁对肾炎家兔的粘附分子及细胞因子表达的影响。方法:以牛血清白蛋白所致家兔急性肾炎模型为观察对象,检测家兔24h尿蛋白含量;ELISA测定血清IL-6;对家兔的肾进行组织形态学观察,免疫组化测定ICAM-1。结果:肾康宁能明显降低实验动物血清IL-6含量,减轻肾小球基底膜(GBM)增厚及炎症细胞浸润,阻止毛细血管内微血栓的形成,降低了急性实验性肾炎模型中ICAM-1阳性细胞表达率。结论:肾康宁通过降低细胞因子IL-6水平和粘附分子ICAM-1的表达,从而抑制免疫细胞的趋化、浸润、活化和增殖,减少自身抗体的分泌使肾小球炎症反应减轻,有效缓解家兔急性实验性肾小球肾炎,使肾功能得以改善。
Objective: To investigate the protective effect of Shenkangning (a complex recipes composed of Chinese medicine) on rabbits with experimental nephritis. Method : BSA were injected to induce rabbits active serological nephritis. Excretion of urinary protein in 24h were tested. IL-6 in blood serum were tested; histomorphology study of renal tissue of models were done. Results: Shenkangning reduced the excretion of urinary protein, decreased expression of ICAM-1 positive cell and IL-6 in the blood serum in the animal models obviously. It may ease the thickening of glomerular bases membrane (GBM) and infiltration of inflammatory cells, meanwhile prevented the formation of micro-thrombus in the capillary of kidney tissue. Conclusions: Shenkangning inhibited chemotaxis, infiltration, activation and proliferation of immune cells and reduced excretion of self-antibody by cutting down the expression of ICAM-1 and the level of IL-6. Thereby Shenkangning eased glomerular inflammation reaction and improved renal function. Shenkangning may play an important significance in prevention and treatment of renal disease.
