血管抑素蛋白质真核胞内表达质粒的构建

摘要 目的:克隆人血管抑素K1-3基因,构建重组酵母胞内表达质粒PHIL-D2/K1-3。方法:从胎儿肝脏组织用反转录-聚合酶链式反应技术(RT-PCR)扩增人血管抑素K1-3目的基因,克隆至酵母胞内表达质粒PHIL-D2中,线性化后转化毕赤酵母GS115菌株,用Sourthern杂交筛选获得阳性重组转化子。结果:用RT-PCR方法成功扩增到750 bp大小的人血管抑素K1-3基因片段,并正向插入毕赤酵母染色体基因组。结论:人血管抑素K1-3基因成功整合到毕赤酵母GS115基因组。
