大肠癌CHD5基因的表达及其与临床病理、预后的关系

THE GENETIC EXPRESSIONS OF CHD5 AND THEIR CORRELATIONS WITH CLINICOPATHOLOGICAL PARAMETERS AND PROGNOSIS OF COLORECTAL CANCER

目的探讨CHD5基因在大肠癌组织和相应正常大肠组织标本中的表达情况;CHD5表达与大肠癌患者临床病理参数和大肠癌生存率和无瘤生存率的关系。方法应用SYBR G reen I实时定量RT-PCR技术检测38例手术切除的大肠癌及相应癌外正常大肠组织标本中CHD5 mRNA的表达,研究CHD5基因检测结果与临床病理参数和预后的关系。结果CHD5 mRNA表达阳性率在癌组织和癌外组织间无差异(100%vs100%,p>0.05)。大肠癌组织CHD5 mRNA的表达水平显著低于癌外正常组织(p<0.05)。大肠癌浸润越深,癌组织CHD5 mRNA的表达量越少。随访病例中,480天的积累生存率为(92.03±4.41)%,无瘤生存率为(77.80±6.97)%。以中位数为界,将癌组织CHD5分为高、低表达组,CHD5 mRNA高、低表达组间无瘤生存率差异无统计学意义(88.89%vs66.67%,p>0.05)。结论CHD5基因与大肠癌有关,其表达水平可能成为判断大肠癌恶性潜能的指标。大肠癌组织CHD5 mRNA的表达水平对患者生存期和无瘤生存期无显著影响。

Objective To investigate the genetic expressions of CHD5 in human colorectal cancer （CRC） and their correlations with clinicopathological parameters ,overall survival rate and disease -free survival rate. Methotis Fresh tumor tissues and mached distal normal colorectal tissues were collected from 38 confirmed CRC patients undergoing surgical operation. The expression levels of CHD5 mRNA were quantified by using SYBR Green I real - time RT - PCR. Results There was no difference of CHD5 mRNA positive rate between CRC tissues and normal tissues （ 100% vs 100%, p 〉 0. 05 ）. The CHD5 mRNA expressions down - regulated significantly in CRC tissues [0. 000 69（0. 000 24,0. 005 8） ] ,compared to those in the corresponding normal colorectal tissues [0. 015（0. 0047, 0. 086） ,p 〈 0.05） ]. The CHD5 mRNA expressions was reversely related to the depths of tumor invasion （p 〈 0. 05 ）. All patients were followed up. The 480 - day cumulative and disease - free survival rates were （92.03±4. 41 ） % and （77. 80±5.97） % ,respectively. When the expression of CHD5 in the CRC tissues were divided into high and low expression groups according to their median values, there was no significant difference in survival rate between high and low expression groups （p 〉 0.05）. Conclusion CHD5 was CRC related genes and might be used as predictive indicators for the malignant potential of CRC ; CHD5 expression might not be a predictor of disease - free survival rate after CRC resection.
