HPLC法测定穿心莲内酯滴丸中穿心莲内酯的含量

Determination of the andrographolide in andrographolide pill by HPLC

目的：探讨测定穿心莲内酯滴丸中穿心莲内酯含量的方法。方法：采用HPLC法测定，使用Hypersil—CN（150mm×4．6mm，5μm）色谱柱，以甲醇：水（40：60）为流动相，流速1．0ml／min，检测波长254nm。结果：穿心莲内酯在20．0～100．0gg／ml范围内有很好的线性关系，其线性方程为A—11484C＋35642（n=5．r=0．9998），回收率为100．53％，RSD=0．39％（n=6）。结论：HPLC法测定穿心莲内酯滴丸中穿心莲内酯线性关系好、准确度高、稳定性好、专属性强，可作为该产品的定量控制方法。

Objective： To establish the determination method for the of andrographolide in andrographolide pill with HPLC. Methods： The HPI.C method was used at a Hypersil-- CN column （ 150mm ×4.6ram, 5μm） as analytical column, with the mobile phase of methanol--water（40：60）. The flow rate was 1.0ml/min,and the detection wave-- length was 254nm. Results： The concentration of andrographolide was linearly related to its peak area in the rang of 20.0-100.0 μg/ml, the linear regression equation was A=11484C＋35642 （n=5, r=0. 9998 ） . The recovery was 100.53% and RSD of 0.39 % （n= 63. Conclusion： The determination of andrographolide in andrographolide pill with HPLC has a good linear relation with the high accurate rate and the good stability, so it can be used for the quantitative analysis.
