摘要
采用乌氏黏度法和凝胶渗透色谱/示差检测器/18角静态激光光散射仪(GPC/RI/MALLS)联用技术,考察不同浓度(0.02%~0.1%)对魔芋葡甘聚糖(KGM)分子在水溶液中行为的影响。分别测定了不同浓度下KGM溶液的黏度值、分子量、多分散性、均方根旋转半径和分子构象。结果表明:随着浓度的升高,黏度值呈指数递增,0.06%为浓度效应拐点;分子量、多分散性、均方根旋转半径测定值随浓度的增加均上升;分子构象先由无规线团分子过渡为球形分子结构,当浓度大于0.06%时,KGM分子构象由球形分子过渡为无规线团分子结构。
Ubbelohde viscosity measurement and gel permeation chromatography/refractive index/18-angle laser light scattering(GPC/RI/MALLS) were used to inspect the effect of concentration(0.02%-0.1%) on konjac glucomannan (KGM) solution behavior. The experiment measured the viscosity, molar mass, polydispersity, root mean square radius of gyration, molecular conformation changes respectively in the different concentration. The results showed that: the values of viscosity increased exponentially with the increase of concentration and the concentration effect of inflection was 0.06%. The molar mass, polydispersity, root mean square radius of gyration, molecular conformation enjoyed rising as well. The molecular conformation was from random coil to globular. When the concentration was greater than 0.06%, the molecular conformation was from globular to random coil.
