响应曲面法优化啤酒废酵母提取β-1,3-葡聚糖工艺的研究 被引量:1

Study on extraction of β-1,3-glucan from waster beer yeast with response surface methodology

摘要 在酵母β-1,3-葡聚糖传统提取工艺上,提出了超声-酶-碱法提取废酵母中β-1,3-葡聚糖的工艺,采用响应曲面试验设计分析了NaOH浓度、温度、用量和时间对β-1,3-葡聚糖品质(得率、纯度与蛋白质含量)的影响,建立了相应的预测模型,并验证了模型的有效性。最优化的条件为NaOH浓度2.05%、温度74℃、用量30.50mL、时间5.7h,在此条件下,β-1,3-葡聚糖成品得率为10.21%,纯度达到88.14%,蛋白质含量为1.19%。 Based on traditional extracting method of β-1,3-glucan from waster beer yeast, an ultrasonic-enzyme-alkali method was developed and introduced in the present paper. The response surface methodology was used to analyze the effects of NaOH concentration, temperature, usage and reaction time on the yield, purity and protein content. The prediction models were also established and verified by the validated data. The result showed that the yield of β-1,3-glucan is 10.21%, polysaccharide content is 88.14% and protein content is 1.19% when the extraction is carried out with 2.05% NaOH concentration, 5.7 h at 74℃ and usage is 30.50 mL.
