X射线诱导多形性胶质母细胞瘤细胞系的凋亡

摘要 辐射诱导肿瘤细胞的凋亡是近年来放射生物学研究的热点,为了进一步探讨辐射诱导肿瘤细胞凋亡的机制,采用光镜、电镜及DNA琼脂糖凝胶电泳法观察X射线诱导多形性胶质母细胞瘤细胞系凋亡的特征。结果表明:①剂量率和分次照射组(10Gy)诱导出典型细胞凋亡的特征(如凋亡小体,DNA梯状带等)。②剂量诱导肿瘤细胞凋亡,高剂量导致肿瘤细胞坏死。③凋亡出现时间因组织细胞类型的不同而各有差异。④射线诱导肿瘤细胞的凋亡以分裂期凋亡为主。 To investigate the mechanism of radiation-induced apoptosis has attracted significant interest to radiobiological research in recent years. Microscope, electronic microscope and DNA agarose gel electrophoresis have been employed to observe the distinct biochemical and morphological features of the radiationinduced apoptosis in human multiform glioblastoma cells (BT325).The results showed that: (1) The classic morphological features of apoptosis(e.g. formation of apoptotic bodies) and the DNA fragmentation ladders on agarose electrophoresis were seen in the low dose's, low doserate's and fractionation group. (2) Apoptosis was induced by low dose X-ray. Necrosis was caused by high dose X-ray.(3) The appearing time of apoptosis was variable in different tissues or cells. (4) Apoptosis induced by X-ray was almost mitosis death. These results provided some new theoretical bases for the clinical radiation treatment and research with glioblastoma multiforme.
