摘要
体外用细胞生长曲线测定法、MTT试验、软琼脂集落形成分析法,及体内对移植性肿瘤实验,研究了海南哥纳香醇甲(GHM10)的抗肿瘤作用,结果表明:GHM10对肿瘤细胞有较强的抑制作用,IC50在2μg·ml-1左右;对正常细胞影响较小,骨髓祖细胞的敏感性则更低;耐药的KB/VCR200细胞及其亲本KB细胞具有相似的敏感性。GHM10对HL60细胞无分化诱导作用。GHM10对实体型肝癌H22小鼠。
The effect of howiinol A (GHM10) on the growth of several cancer and normal cells were studied, using the methods of cell growth curve determination, MTT test and softagar colony formation assay. The results showed that GHM10 exhibited potent inhibitory effect on cancer cells with an IC50 of 2 μg·ml-1 approximately. Normal cells, especially bone marrow progenitor cells, were shown to be less sensitive to GHM10. A drugresistant cell line, KB/VCR200, was found to be as sensitive as its parental KB cells. GHM10 did not induce HL60 cell differentiation. In murine transplantable tumors, GHM10 was found to have significant therapeutic effect on mice bearing solid tumor hepatoma H22, Lewis lung carcinoma and S180 ascites.
