摘要
目的探讨聚乳酸-三亚甲基碳酸酯/胶质细胞源性神经营养因子[P(LA-TMC)/GDNF]复合导管中促进大白鼠脊髓再生的GDNF的合适含量。方法Wistar雌性大白鼠60只,随机分为6组,每组10只。各组采用P(LA-TMC)/GDNF复合导管中的GDNF含量分别为150 u3、00 u4、50 u5、50 u和700 u8、50 u。将每组动物的脊髓造成4 mm缺损的间隙,分别以不同GDNF含量的[P(LA-TMC)]/GDNF复合导管桥接缺损。术后2个月检测比较各组脊髓的再生情况。结果GDNF含量为450 u和550 u的P(LA-TMC)/GDNF复合导管桥接组的脊髓再生情况显著优于其他4组(P<0.01或P<0.05),而该2组间的差异无统计学意义(P>0.05)。其中GDNF含量为150 u和800 u组的神经再生情况最差。结论P(LA-TMC)/GDNF复合导管中GDNF的含量为450-550 u左右,对促进大白鼠脊髓再生最为合适。
Objective To analyse the suitable dose of in the P(LA-TMC)/GDNF on restoration of rat spinal defect.Methods 60 female Wisar rtar rats were randomly assigned into six groups with 8 each. The doses of GDNF in the P(LA-TMC)/GDNF conduit were 150 u,350 u,450 u,550 u,and 850 u respectively. The spinal of the animals was transected to create a 4 mm gap and then bridged with P(LA-TMC)/GDNF compound conduit containing different doses of GDNF according to group assignment. Spinal regeneration was evaluated at 2 months following the operation by potential plane test, comparing by basso, beattie and bresnahan (BBB) score, comparing by myelinated nerve fiber count and axon diameter among the groups. Results Nerve regeneration in the groups containing 450 u and 550 u GDNF was significantly superior to those in the other four groups. There was no difference between parametersof 450 u GDNF group and 550 u GDNF group. Among the 6 groups nerve regeneration was most unfavorable in the 850 u GDNF and 150 u GDNF group. Conclusion The suitable dose of GDNF in P( LA-TMC)/GDNF compound conduit is around 450 - 550 u. Higher or lower concentrationsof GDNF could inhibit nerve regeneration.
