摘要
为筛选高转移肺癌中差异表达的基因,探究相关基因的生物学功能.利用荧光差异显示技术(DD-PCR)获得差异片段,对其进行克隆和序列分析.利用定量PCR检测验证该基因表达的差异性,对它的结构功能进行预测,并进行原核和真核表达.通过DD-PCR得到差异片段,经测序分析表明:C3orf1基因在高转移肺癌中的表达量显著高于肺癌细胞.该基因在原核和真核中都得到了表达,可能与高转移肺癌转移相关.
To further define molecular mechanism involved in metastasis process and screen out the genes expressed differentially in lung cancer, the highly expressed fragments were cloned and sequenced. The gene was expressed in the prokaryotic cell and eukaryotic cell. One of the differentially expressed genes, which was called C3orf1, showed high different expression within the six tested cancer cells. The result of the DD-PCR indicates that the gene C3orf1 is expressed highly in the metastasis cells. Moreover, the gene is also expressed in the prokaryotic cell and eukaryotic cell successfully. The gene C3orf1 is over-expressed in the lung cancer 95D cells, and it may play an important role in stimulating the cells growth.
