摘要
以狭鳕鱼皮为原料,根据Box-Behnken的中心组合试验设计原理,在单因素试验的基础上采用响应面分析法,建立了微波辅助提取狭鳕鱼皮明胶的二次多项数学模型,并以明胶提取率为响应值和等高图,考察了NaOH质量分数、H2SO4质量分数、微波功率和微波处理时间对鱼皮明胶提取效果的影响。结果表明:NaOH质量分数、H2SO4质量分数、微波功率、微波处理时间对鱼皮明胶提取率都有显著影响,微波辅助提取狭鳕鱼皮明胶的最佳的工艺条件为,NaOH质量分数0.09%、H2SO4质量分数0.21%、微波功率578W、处理时间16.4min,鱼皮明胶的提取率达到极大值。此条件下鱼皮明胶提取率的实测值为20.65%。与常规加热法提胶相比,微波辅助法提取率提高了3.1%,时间缩短了97.8%。
The microwave - assisted extraction process of fishskin gelatin from Alaska Pollock was investigated. On the basis of single factor tests and the Box - Behnken center united experimental design principle, the method of response surface analysis was adopted to build a second order quadratic equation for microwave - assisted extraction of fishskin gelatin. Moreover, response surface and contour were graphed with the gelatin yield as the response value, effects of NaOH concentration, H2SO4 concentration, microwave power and reaction time on the extraction effect were explored. The result showed that the optimum extraction condition was obtained as follows: NaOH concentration of 0.09% , H2SO4 concentration of 0.21% , microwave power of 578W, reaction time of 16.4min. Under the optimum condition, the measured gelatin yield was 20.65%. Compared with the traditional heating extraction method, the yield was increased by 3.1% , extraction time was increased by 97.8 %.
