新生小鼠子宫内膜上皮层体外分离研究

摘要 为探讨新的体外分离培养新生小鼠子宫内膜上皮细胞的方法,尝试先体外分离新生小鼠子宫内膜上皮层,再从分离后的上皮层分离培养上皮细胞的技术路线,取新生3-4日龄的小白鼠子宫角,4℃下置于1%胰蛋白酶/0.02%EDTA消化液中温和消化90min,将巴斯德吸管的管径拉成等于或略小于子宫角直径大小,采用吹打的方法机械分离小鼠子宫内膜上皮层与基质层.结果表明:采用胰蛋白酶温和消化子宫角结合机械分离法可成功体外分离新生小鼠子宫角的子宫内膜上皮层,使先体外分离新生小鼠子宫内膜上皮层,再从分离后的上皮层分离培养上皮细胞的技术路线成为可能. In order to find a new way to separate and culture neonatal mouse in vitro, 1% typsin -0.02% EDTA solution cells of endometrial epithelium from uterus horns of is used to gently digest uterus horns get from 3 to 4 days mice under 4 ℃ for 90 minutes, then these uterus horns are blow out though the bore of a Pasteur pipette tip which equal or slightly smaller than the cross- sectional diameter of the uterine piece till the layers of endometrial epithelium and the stroma separated. The results show that the method of separating neonatal mouse endometrial epithelium is effective and make the technique that separate and culture cells of endometrial epithelium from isolated endometrial epithelium become true.
