摘要
【目的14g讨病毒性心肌炎(VMC)患儿T淋巴细胞CD40和RANTES表达对其自身凋亡的调控机制。【方法】选择VMC患儿和正常儿童各25例,采用ELISA检测培养外周血T淋巴细胞RANTES浓度,FITC单标和AnnexinV/PI双标流式细胞仪分别定量检测两组外周血T淋巴细胞CD40表达及凋亡百分率。【结果】VMC组CD40分子表达水平和RANTES浓度与对照组比较均有明显升高(P〈0.01),且两者在VCM组呈明显正相关(r=0.295;P〈0.01);VMC组RANTES浓度和T淋巴细胞凋亡率与对照组比较均有明显升高(P〈O.01),且两者在VCM组明显正相关(r=0.467;P〈0.01)。【结论】VMC患儿T淋巴细胞膜上CD40可能通过活化CD40-CD40L信号通路参与了对T淋巴细胞RANTES分泌的调控,从而进一步调控了T淋巴细胞的凋亡。
[Objective]To investigate the mechanism in T-lymphocyte apoptosis of viral myocarditis (VMC) regulated by the expression of CD40 and RANTES. [Methods]Twenty five VMC cases and matched normal children as control were chosen. RANTES concentration in cultured peripheral blood T-lymphocytes was detected by ELISA. The peripheral blood T-lymphocyte percentages of CD40 expression and apoptosis in two groups were measured by standard single-FITC and Annexin V/PI double-labeled flow cytometry. [Re suits] The CD40 expression level as well as the concentration of RANTES in VMC were significantly higher compared with the control group( P 〈0.01) and were significantly positive correlation(r=0. 295; P 〈0.01). Peripheral blood T-lymphocyte percentage of apoptosis as well as the concentration of RANTES in VMC were significantly higher compared with the control group( P 〈0.01) and were significantly positive correlation(r= 0. 467; P〈0.01). [Conclusion]RANTES secretion in T-lymphocyte of VMC is maybe regulated by the ex- pression of the CD40 on T-lymphocyte membrane through CD40-CD40L signaling pathway, thereby further regulation in T lymphocyte apoptosis is controlled.
