白藜芦醇对大鼠急性脊髓损伤后运动功能及细胞凋亡的影响 被引量:6

摘要 目的探讨白藜芦醇(Res)对大鼠急性脊髓损伤后运动功能、组织病理学及神经细胞凋亡的影响。方法本文采用Allen′s脊髓打击模型,将24只SD大鼠,随机分为3组,每组8只。A组:假手术组;B组:脊髓打击伤组;C组:Res处理组。术后分别观察24 h4、8 h、72 h BBB评分变化;72 h处死动物取材,HE、Nissl染色观察脊髓组织病理学变化;原位末端脱氧核糖核苷酸转移酶介导dUTP标记技术检测神经细胞凋亡情况。结果BBB评分提示3个时相点均有所改善,且C组均高于B组,呈显著性差异(P<0.05);组织病理组织切片观察显示C组结构破坏明显轻于B组;TUNEL阳性神经细胞均有表达,C组明显少于B组。结论Res能明显促使脊髓损伤后的大鼠后肢的神经运动功能恢复,保护神经结构破坏和抑制神经细胞凋亡。 To investigate the effect of resveratrol (Res) on motor function, histopathology and neuronal cells apoptosis after spinal cord injury in rats. Methods: 24 SD rats were randomly divided into 3 groups. A group: sham-operation group; B group :spinal cord injury group; C group Res group. Animal model of acute spinal cord was induced through Allen's method. In RES group, At last, the hindlimb motor function in the spinal cord were observed at the 24, 48 and 72 h after modeling. The samples were taken from the spinal cord injury sites at 72 hours after the injury and tested by HE,Nissl staining for histopathological a and by TUNEL method for cell apoptosis. Resuhs:The hindlimb motor function of C group and B group became better at the 24hours,48 hours,72 hours, and there was obvious difference( P 〈 0.05 ) ; Histopathological observation showed that tissue damage to the structure of C group significantly lighter than B group; The apoptotic cells positive of TUNEL were detectable in beth three experimental groups. In the C group, the number of apeptotic cells was less and neuronal function of the spinal cord better as compared with those in the B group. Conclusion: Res significantly to spinal cord injury in rats after hindlimb motor function recovery of the nerve to protect the neural structure of nerve cell damage and inhibition of apoptosis.
