摘要
针对现有细胞电融合仪的不足,研究开发了一种基于微系统技术的高效自动细胞电融合仪。该仪器将MEMS工艺加工的电融合芯片与电融合信号发生器集成为一体,通过精确的结构设计和信号产生来控制细胞的位置。由于芯片的结构特点,使融合控制电极间距减小,从而降低了对融合电压的要求。融合装置可选用多种不同尺寸规格的电极,而且,细胞融合进程可以直接进行显微观察和操作。融合实验表明:该装置可以提高细胞融合效率,融合率达到40%。
Aimed at the deficiency in the existing cell-elctrofusion devices, a microsystem technology-based highefficiency and automatic cell-electrofusion device is developed. MEMS technique fabricated cell-electrofusion chip is integrated with signal generator, in which precisely designed microstructure and signal are used for locating cells. In this chip, small distance between one pair of microelectrodes reduces required voltage. Many differentsize microelectrodes can be chosen in this device. Cell fusion can also be observed and manipulated under microscope. According to the result in the on-chip cell-fusion experiments, up to 40 % fusion efficiency is obtained.
