大孔吸附树脂对淫羊藿中总黄酮及淫羊藿苷吸附纯化的研究 被引量:12

摘要 目的:探讨大孔吸附树脂对淫羊藿中总黄酮及淫羊藿苷吸附纯化的初步研究。方法:采用大孔树脂法分离纯化淫羊藿中总黄酮成分及淫羊藿苷,并应用紫外分光光度法和高效液相色谱法,分别以淫羊藿总黄酮和淫羊藿苷为考核指标,筛选大孔吸附树脂类型,并对洗脱溶剂的选择、树脂柱径高比、洗脱流速、药材与树脂用量比、洗脱剂用量等进行考察。结果:以AB-8为最优型号树脂用于分离纯化淫羊藿中总黄酮和淫羊藿苷,以70%乙醇,树脂柱径高比为1∶3,洗脱流速6BV/h,药材树脂用量比1∶3为最佳工艺。结论:采用大孔树脂法分离纯化淫羊藿中总黄酮及淫羊藿苷,并应用高效液相色谱法测定淫羊藿苷含量,紫外分光光度法测定总黄酮含量,证明该法具有简便、快速、准确等优点。 Objective: To collect and purify of total flavonoids and icariin on Herba Epirnedii. by macroprous resin. Methods: To determine the best technology of collecting and purifing the total flavonoids and icariin in Herba Epimedii. by macroprous resin while total flavonoids was as adopted as standard sbstance by ultraviolet spectrophotometry and icariin was as adopted as standard sbstance by HPLC. Results: AB-8 was proved to be the most suitable one to purify the total flavonoids and icariin in the experiment. The best technology was determined as follows: extracted solvent was 70% ethanol, the ratio of diameter to hight was 1:3, elution rate was 6BV/h, the weight ratio of material to resin was 1:3. Conclusion: It is suitable to collect and purify the total flavonoids and icariin by macroprous resin and determine the content of total flavonoids by ultraviolet spectrophotometry, icariin by HPLC in Herba Epimedii.
