摘要
目的:观察高糖对不同状态下乳鼠心肌细胞肿瘤坏死因子-α(TNF-α)基因表达的影响,探讨TNF-α在糖尿病心肌病的非特异性炎症中的作用机制。方法:体外培养乳鼠心肌细胞,分别用不同浓度的葡萄糖(0、20、30、50mmol/L)干预正常心肌细胞、胰岛素抵抗心肌细胞,采用逆转录聚合酶链反应检测细胞TNF-αmRNA的表达水平,并在透射电镜下观察心肌细胞结构的变化。结果:(1)在50mmol/L葡萄糖作用下,正常心肌细胞TNF-αmRNA表达(0.58±0.03)明显高于20mmol/L(0.27±0.02)、30mmol/L(0.29±0.05)葡萄糖作用的正常心肌细胞,而后两组之间比较无统计学差异。(2)胰岛素抵抗心肌细胞在0、20、30、50mmol/L葡萄糖作用下,各组心肌细胞TNF-αmRNA表达均有显著性差异(P<0.05),并随着葡萄糖浓度的提高,TNF-αmRNA表达越高;在相同浓度葡萄糖作用下,胰岛素抵抗心肌细胞TNF-αmRNA表达明显高于正常心肌细胞。(3)在高浓度葡萄糖作用下,正常心肌细胞和胰岛素抵抗心肌细胞出现线粒体肿胀,髓样结构形成,胞内脂滴增多等细胞结构损伤性改变。结论:高糖作用下胰岛素抵抗心肌细胞TNF-αmRNA表达明显增加,引起胞内脂滴增多等细胞结构变化。
Objective:To investigate the effects of high-concentration glucose on the expression of tumor necrosis faetor-α (TNF-α) mRNA in cultured rat cardiomyocytes and to analyze the pathogenesis of TNF-α as the inflammatory factor in diabetic cardiomyopathy. Methods:Cultured prima- ry rat cardiomyocytes were assigned to two groups: the normal cardiomyocyte group and the insulinresistent cardiomyocyte group. The two groups of cardiomyocytes were both incubated with various concentrations of glucose (0, 20, 30, 50mmol/L). The expression of TNF-α mRNA was measured by RT-PCR. The changes of eellstructure were observed through transmission eleetromieroseope. Results: (1)The difference of the expression of TNF-α mRNA between the normal cardiomyocytes treated with 50 mmol/L glucose(0.58 ± 0.03)and the normal cardiomyoeytes treated with 20 mmol/ L glucose(0.27 ± 0.02), 30 mmol/L glucose(0.29 ± 0.05)was significant(P(0.05) ,the latter two were not different significantly compared with each other (P) 0.05). (2) The expression of TNF-α mRNA of the insulin-resistent cardiomyocytes with 0, 20, 30, and 50mmol/L glucose were different significantly compared with each other(P〈0. 05). The expression of TNF-α mRNA was increased with the increasing concentration of the glucose to some extent. The expression of TNF-α mRNA of the insulin-resistent cardiomyocytes was increased compared with normal cardiomyocytes surrounded by the same concentration of the glucose. (3)Swelled mitochondria, formed myelir figure, increased lipid were observed through transmission electromicroscope in normal cardiomyocytes and the insulinresistent cardiomyocytes. Conclusion: High-concentration glucose could increase the expression of TNF-α mRNA and induce the changes of cellstructure such as increased lipid in normal cardiomyocyte and insulin-resistent eardiomyocytes.
