摘要
利用正硅酸乙酯与壳聚糖通过溶胶-凝胶反应制备了壳聚糖/SiO2杂化膜,用来固定糖化酶。对固定化酶的酶学性质进行了研究,结果表明:SiO2含量为4%的杂化膜相对酶活最高,达到了77.91%。固定化酶的最适作用温度为50℃,最适作用pH为5.2,固定化酶的耐受热性及pH稳定性较游离酶有所提高。
The chitosan/silica hybrid membrane was prepared with chitosan and TEOS by the method of sol - gel. h was used to immobilize glucoamylase. The enzymatic properties of immobilized enzyme was studied and the results showed that the highest relative enzyme activity of the hybrid membrane was 77.91%. The optimal reaction temperature of the immobilized enzyme was 50℃ and its optimal pH was 5.2. The temperature and pH stability of immobilized enzyme were higher than free enzyme.
