摘要
目的:研究红景天苷对胸主动脉环的舒张作用并探讨其可能的机制。方法:采用兔胸主动脉环,观察红景天苷对胸主动脉环张力的影响。取原代培养的牛胸主动脉内皮细胞,观察红景天苷对内皮细胞一氧化氮(NO)和总一氧化氮合酶(tNOS)的影响。结果:红景天苷对正常胸主动脉环没有明显的舒张作用;对1μmo.lL-1去甲肾上腺素(NE)和60 mmo.lL-1KCl引起的胸主动脉环收缩有舒张作用,去除内皮后的胸主动脉环舒张功能降低;左旋硝基精氨酸甲酯(L-NAME)可明显抑制红景天苷对胸主动脉环的舒张作用;红景天苷能增加内皮细胞NO释放和细胞裂解液中tNOS活性。结论:红景天苷对预收缩的胸主动脉环有明显内皮依赖性和非内皮依赖性舒张作用,内皮依赖性舒血管作用与NO途径有关。
Objective: To investigate the vasodilatation effect of salidroside on thoracic aorta and its related mechanism.Methods: The isolated rabbit aortic rings were used to observe the effect of salidroside in vitro.The effects of salidroside on NO and tNOS were observed in aortic endothelial cells.Results: Salidroside had no significant relaxation effect on normal thoracic aorta rings,but relaxed thoracic aorta rings pretreated by norepinephrine(NE,1 μmol·L^-1) or KCl(60 mmol·L^-1),and the relaxation in endothelium-denuded rings was reduced.NG-nitro-L-arginine methylester(L-NAME) significantly inhibited the relaxation effects of salidroside on thoracic aortic rings.Salidroside increased the level of NO and the activity of tNOS.Conclusion: Salidroside induced both endothelium-dependent and endothelium-independent relaxation in thoracic aorta rings pretreated by norepinephrine.Moreover,NO pathway might be involved in the endothelium-dependent relaxation.
