摘要
目的观察HSP70、NMP238在放疗敏感性不同宫颈癌组织中的表达及与宫颈癌放疗敏感性的关系。方法收集进行治疗前早期宫颈癌组织标本,病理诊断均为中分化鳞癌,置于-80℃超低温冰箱保存。在进行放疗后,根据WHO实体瘤疗效判断标准,将收集的宫颈癌组织标本10例分为高敏感组(5例)和低敏感组(5例)。提取组织总蛋白,进行双向凝胶电泳和质谱分析鉴定差异表达蛋白,分别应用W estern b lot和免疫组织化学方法检测组织中蛋白的表达。结果建立了分辨率高、重复性好的宫颈癌放疗高敏感组和低敏感组的双向凝胶电泳图谱。差异表达蛋白经鉴定得到低敏感组高表达蛋白HSP70和NMP238。W estern b lot检测结果显示,HSP70和NMP238在低敏感组中的表达强于高敏感组。免疫组化结果显示,HSP70和NMP238在低敏感组中的表达强度及表达率高于高敏感组。结论HSP70和NMP238的表达与宫颈癌放疗敏感性有关。
Objective To observe HSP70 and NMP238 expression in cervical carcinoma tissues with different radiotherapy sensitivity and its relation to radiotherapy sensitivity of cervical carcinoma. Methods The fresh carcinoma tissues were collected from untreated cervical carcinoma patients and preserved in the -80℃ refrigerator. The tissues were classified into two groups: high sensitivity group (HS) and low sensitivity group( IS), according to their response to radiotherapy. In the first part of the experiment, protein separation was performed using two-dimensional gel electrophoresis (2-DE) and matrix assisted laser desorption/ianization time-of-flight mass spectrometry(MALDI-TOF-MS). The proteins were identified by data searching from the Mascot-database. Differential expression of proteins were assayed by Western Blot and immunohistochemical method. Results Most of the gels were clear and successfully and reproductively analyzed. HSP70 and NMP238 expression was higher in LS group than in HS group. HSP70 and NMP238 in LS group were further confirmed by Western blot and immunohistochenmical method. Conclusions HSP70 and NMP238 expression is associated with radiotherapy sensitivity of cervical carcinoma.
