摘要
目的研究饮水型氟中毒对脊髓神经细胞的作用机制,特别是与神经细胞凋亡的关系。方法60只Wistar大鼠随机分为两组,实验组喂以预配的高浓度氟化钠水建立饮水型氟中毒大鼠模型,采用原位末端标记法(TUNEL)和免疫组化半光氨酸蛋白酶(Caspase-3)测定评价饮水型氟中毒大鼠模型脊髓神经细胞的凋亡情况。结果TUNEL结果显示,饮水型氟中毒大鼠模型脊髓中可见明显的细胞核棕染的凋亡神经细胞,Caspase-3检测,实验组相比对照组呈现过表达。结论饮水型氟中毒大鼠模型的脊髓神经细胞有凋亡倾向,Caspase-3的参与和过表达可能是饮水型氟中毒导致神经细胞发生凋亡而致脊髓损伤的原因之一。
Objective To study the expression of neurocyte apoptosis and the changes of Caspase-3 in rats with fluorosis caused by drinking fluorinated water. Methods 60 Wistar rats were separated randomly into 2 groups,rats in one group feeded by prepared high concentration NaF water to make fluorosis model and classified as study group;others feeded by distilled water as control group. Terminal deoxyribo-nucleotide transferase-mediated UTP-biotin nick end labeling (TUNEL) and immunohistochemical investment through Caspase-3 test were used to detect cytologic changes of neurocyte in spinal cord after the fluorosis model had been made. Results Apoptosis neurocyte were detected significantly in gray matter of spinal cord of fluorosis rats in study group compared with those in control group through TUNEL. Caspase-3 staining results also show overexpression in rats with fluorosis caused by drinking fluorinated water in study group. Conclusion There is a tendency of apoptosis in spinal neurocyte in rats with fluorosis caused by drinking fluorinated water, Caspase -3 probably plays an important role in the mechanism of apoptosis caused by drinking fluorinated water.
