摘要 目的:探讨特异性微小RNA抑制剂对胃癌细胞增殖的影响。方法:设计并合成4种微小RNA(miR-17、miR-21、miR-106a和miR-421)的2-甲氧修饰的RNA寡核苷酸(微小RNA抑制剂),用脂质体分别转染到SGC-7901和MGC-803胃癌细胞,然后用实时定量逆转录-聚合酶链反应技术检测微小RNA的表达情况,最后用MTT方法检测胃癌细胞的增殖情况。结果:4种微小RNA抑制剂均有效抑制了SGC-7901和MGC-803细胞中相应微小RNA的表达;除miR-21抑制剂未见抑制胃癌细胞的增殖外,其它3种微小RNA抑制剂均以剂量依赖方式抑制胃癌细胞的增殖且对SGC-7901的效果优于MGC-803。结论:微小RNA的特异性抑制剂能有效抑制胃癌细胞的增殖,采用这种技术为研究胃癌的发病机制提供了新方法。 AIM: To investigate the effects of miRNA - specific inhibitors on proliferation of gastric cancer cells. METHODS : The specific 2' - methoxy - modified RNA oligonucleotides ( miRNA inhibitors ) of miR - 17, miR - 21, miR - 106a, and miR -421 were designed, synthesized and transfected into SGC -7901 and MGC -803 gastric canc- er ceils by liposome. The expression of miRNAs in cells was detected by quantitative real - time reverse transcriptase - pol- ymerase chain reaction. Finally, the cell proliferation was analyzed by MTT assay. RESULTS : Four miRNA - specific in- hibitors significantly inhibited the expression of corresponding miRNAs in SGC - 7901 and MGC - 803 cells, miRNA inhib- itors of miR - 17, miR - 106a, and miR -421 showed the inhibitory effect of proliferation on gastric cancer cells in a dose - dependent manner. The growth inhibitory effect on SGC - 7901 cells was better than that on MGC - 803 cells. CON- CLUSION: The miRNA -specific inhibitors effectively inhibit the proliferation of gastric cancer cells and provide a new method for investigating the pathogenesis of gastric cancer.
