摘要 目的:从能够降解黄曲霉毒素B1的嗜麦芽窄食单胞菌中提取主要降解成分,并对其降解作用进行研究。方法:采用硫酸铵沉淀法对发酵液进行酶的提取,并对酶的提取条件进行优化实验。结果:确定种子培养基培养12h,发酵24h后,采用65%硫酸铵沉淀后的酶活性最大,对黄曲霉毒素B1的降解率达到80.25%。结论:降解黄曲霉毒素B1的菌株于40℃下种子培养12h,发酵24h,经过65%硫酸铵沉淀得到粗酶提取液,其蛋白含量为532.9μg/mL,与黄曲霉毒素B1反应72h后酶活最大,对毒素的降解率可达到81.23%,并且经过全波长扫描得到此酶在240~267nm有最大吸收峰。 Objective:The aim of this text was mainly to extract the main ingredients from the Stenotrophomonas sp. which can degrade aflatoxin B1 . Method : Using ammonium sulfate precipitation method for enzymatic extraction, and optimize the conditions of enzyme degraded reaction. Result: Use 65% ammonium sulfate precipitation after 24h fermentation,the degradation ratio of AFB, reached 80.25% ,which was obviously higher than others and the enzyme had the greatest activity. Conclusion: The degraded microorganisms fermentation at 40~0 for 24-h, its protein content was 532.9μg/mL after 65% ammonium sulfate precipitation.The degradation ratio of AFB, reached 81.23% after 72h reaction.This enzyme had absorption peaks in the 240-267nm through a full-wavelength scan.
