几种单糖、寡糖TLC条件的确立及草石蚕寡糖中水苏糖含量测定 被引量:8

摘要 目的:建立薄层色谱(TLC)分析几种单糖、寡糖的有效方法,并测定草石蚕寡糖中水苏糖含量。方法:以果糖、蔗糖、棉子糖和水苏糖为研究对象,从斑点分离度、斑点集中与否、有无拖尾现象及展开时间几方面探讨其在不同体系下的薄层色谱行为;通过薄层色谱扫描(TLCS)测定草石蚕寡糖中水苏糖含量。结果:确立适合的展开系统为乙酸乙酯/甲醇/水/冰乙酸(11.5:3.2:2:3.2),合适的显色剂为茴香醛-浓硫酸显色剂;草石蚕寡糖中水苏糖含量为69.3%。 Purpose: To establish an effective TLC method for the analysis of several monosaccharides and oligosaccharides and determinate the content of stachyose in in oligosaccharide from Stachys seiboibi Miq. Methods:TLC with different developing systems was applied to the analysis of fructose, sucrose, raffinose and stachyose, related to the degree of isolated spots, the spots focusing on it or not, tailing phenomenon and expanding time. TLCS was applied to determinate the content of stachyose in oligosaccharide from Stachys seiboibi Miq.Results:A mixed solution composed of ethyl acetate, methanol, water and glacial acetic acid in a ratio of ethyl acetate: methanol:water: glacial acetic acid = 11.5:3.2:2:3.2 by volume was used as development reagent.A solution containing anisaldehyde and concentrated sulfuric acid was used as color reagent.The content of stachyose in oligosaccharide from Stachys seiboibi Miq was 69.3%.
