丁型肝炎病毒RNA巢式逆转录PCR检测

Detection of hepatitis D virus RNA in serum by using reverse transcription nested polymerase chain reaction

为了提高丁型肝炎病毒的实验室诊断水平，建立一个高敏感性和特异性的丁型肝炎病毒的检测方法。根据ＧＥＮＢＡＮＫ中ＨＤＶ的核酸序列设计出巢式引物，采用逆转录巢式ＰＣＲ方法对４０例肝炎患者进行检测。结果：２０例丁型肝炎病毒抗原阳性的ＨＤＶＲＮＡ均为阳性，１０例丁型肝炎病毒抗体阳性中ＨＤＶＲＮＡ阳性６例，１０例单纯ＨＢｓＡｇ、ＨＢｅＡｇ和抗ＨＢｃ阳性的均未检出ＨＤＶＲＮＡ。

In order to improve the diagnostic rate of hepatitis D virus (HDV),a highly sensitive and specific method has been established in which a nested primer was designed according to HDV sequences from Genbank.Forty serum samples from patients with hepatitis were examined by using the reverse transcription nested polymerase chain reaction (RT nested PCR).The results revealed that HDV RNA was detected in 20/20 of the positive serum samples for HDAg and 6/10 of the positive samples for anti HD(negative HDAg),while HDV RNA was negative in 10 positive samples for HBsAg,HBeAg and Anti HBc.It is concluded that RT nested PCR may be useful in routine detection of HDV RNA.
