蛋白酪氨酸磷酸酶SHP-2在前列腺肿瘤组织中的表达 被引量:1

摘要 目的:探讨磷酸酪氨酸和含C-src同源序列的酪氨酸磷酸酶SHP-2在前列腺肿瘤组织中的表达。方法:采用免疫组化En Vision二步法检测10例正常前列腺组织、30例良性前列腺增生组织(BPH)、20例前列腺上皮内瘤(PIN)、20例高分化前列腺癌(Pca)和10例低分化前列腺癌组织中磷酸酪氨酸和SHP-2的表达。结果:磷酸酪氨酸在正常前列腺中基本不表达,在BPH和PIN组织中主要分布在分泌细胞和基底细胞的胞浆中,呈细颗粒状,分泌细胞部分胞膜有染色,BPH组织以低表达为主,而PIN组织中的表达有所增强,在Pca组织中,主要分布于肿瘤细胞的胞浆中,并呈粗颗粒状表达,部分分泌细胞胞膜有表达。磷酸酪氨酸在各组的平均染色指数分别为0.2、1.2、1.9、2.2和2.4。SHP-2在正常组中主要表达在基底和上皮细胞的胞浆中,少数在胞核中表达,在BPH组中在基底和上皮细胞的胞浆和胞核中均有表达,在PIN、高分化Pca和低分化Pca组中SHP-2的表达主要在细胞核内,少数在胞浆中表达,SHP-2在各组的平均染色指数分别为0.4、1.7、2.1、2.2和2.6。结论:在前列腺组织细胞增殖、分化及转化过程中,存在磷酸酪氨酸和SHP-2相关的信号传导的改变。磷酸酪氨酸的异常表达和SHP-2异常激活、表达部位的改变可能是导致前列腺癌变的重要机制。 Objective:To explore the expression of phosphorylated tyrosine and tyrosine phosphatase SHP -2 in prostate cancer. Methods:With envison two- step method, the expression of phosphorylated tyrosine and SHP-2 was detetecd in 10 cases of normal prostate tissue, 30 of BPH, 20 of PIN, 20 of high differential Pca and 20 of low differential Pca. Results : No - expression of P - Tyr were shown in normal prostate tissues. The expression of P - Tyr strengthened according to the severity of the illness in BPH, PIN and Pca. The expression mainly distributed in the cytoplasm of secretive cells and basal cells. Some P - tyr expression was also observed on the membrane of secretive cells. The average dying index of P - Tyr in each group was 0. 2,1.2, 1. 9,2. 2 and 2.4. The expression of SHP - 2 in normal group mainly distributed in the cytoplasm of secretive cells and basal cells, and a little part in the nucleu. In BPH, it distributed equally in the plasm and nucleu. In PIN, high differential Pca and low differential Pca, SHP - 2 expressed mainly in nucleu. The average dying index of SHP - 2 in each group was 0.4, 1. 7,2. 1,2. 2 and 2. 6. Conclusion:There is transformation in signal transduction relation with phosphorylated tyrosine and SHP- 2 in the progress of prostate cell proliferation, differentiation and malignancy. The abnormal expression of phosphorylated tyrosine, the abnormal activation and abnormal distribution of SHP -2 might induce prostate cancer.
