摘要
目的:建立一种适用于生化自动分析仪动力学测定磷酸甘油酸变位酶(phosphoglyceromutase,PGAM)的方法并评价此方法的可靠性。为进一步探讨人体血清PGAM活性在急性心肌梗死和脑出血等疾病中的早期作用建立良好的方法基础。方法:PGAM活性测定采用3-磷酸甘油酸为底物,经烯醇化酶等一系列偶联,以340 nm波长在自动分析仪上测定,根据NADH吸光度变化可求出PGAM总活性。结果:通过一系列参数的测定建立了良好的能适应生化自动分析仪的PGAM活性测定方法。结论:酶偶联法操作简单、测定快速、稳定性好,灵敏度和精确度高,抗干扰性强,可在全自动生化分析仪上应用;实验证明PGAM对于早期诊断心肌梗死,判断预后有一定价值;另一方面,脑出血时PGAM变化特点是:脑出血发作后,PGAM的活性在2 h内即可上升,然后下降,与心肌梗塞有明显不同,所以测定PGAM活性有助于临床上的鉴别诊断。
Objective: To establish an automatic biochemical analyzer PGAM dynamics of the method and to evaluate the reliability of this method. To further explore human serum PGAM activity in acute myocardial infarction and cerebral hemorrhage, and other diseases in the early role of a good basis for the method. Methods:The activity was determined by 3 - phosphoglycerate as substrate, the enolase and a series of coupling to 340 nm wavelength automatic analyzer in the determination of total activity can be derived. Results: Through a series of parameters to establish a good automatic biochemical analyzer to the PGAM assay. Conclusion:Coupling method is simple and of fast - determination, good stability, high accuracy and sensitivity, and strong anti - interference, in the automatic biochemical analyzer application.
