摘要
目的研究bFGF、Bax及细胞凋亡在胎膜早破发生发展中的作用。方法选择行剖宫产分娩的初产妇90例,其中胎膜早破者(PROM组)45例,足月完好胎膜者(对照组)45例,采集胎膜组织。①采用免疫组化法检测胎膜组织中bFGF和Bax蛋白的表达;②采用原位末端TUNEL法检测胎膜组织中的凋亡细胞。结果①bFGF和Bax在PROM组中的表达水平显著高于对照组(P<0.05);②PROM组凋亡细胞较正常对照组明显增加(P<0.05)。结论①Bax在PROM患者胎膜组织中呈高表达,提示Bax/Bc l-2比值失衡可能是诱发PROM的细胞凋亡途径之一;②内源性bFGF可能参与胎膜损伤后的自身应激性修复过程,因而有可能作为临床上PROM的治疗手段。
Objective To research the expressions of bFGF and Bax in the pathogenetic mechanisms of premature rupture of the membranes. Methods 45 normal tissues of fetal membranes and 45 tissues of premature rupture of the fetal membranes (PROM) were collected. The expressions of bFGF and Bax in the fetal membrane were assayed by immunohistochemical method. The apoptosis in the fetal membrane were assayed by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling (TUNEL). Results ①The expression levels of bFGF and Bax in the amniochorion with PROM were higher than those in control group ( P 〈 0.05 ). ②The apoptosis in the PROM group was higher than that of in the control group (P 〈 0.05). Conclusion ① The increased expression of Bax in PROM, which induce apoptosis by breaking the balance of heterodimeric Bax/Bcl - 2 complexes, may be one of the important mechanisms in the pathogenesis of PROM. ② suggests that intrinsic bFGF may play a role of PROM treatment for PROM. The increased expression of bFGF in the PROM repairing, which might be a potential therapeutic treatment for PROM.
