白皮松成熟胚不定芽诱导分化技术的优化 被引量:5

摘要 以白皮松成熟胚为外植体进行不定芽诱导分化技术的优化,以此建立白皮松的再生体系。探讨了基本培养基、激素配比、糖浓度和不同摆放方式及不定芽继代增殖等问题。结果表明:成熟胚不定芽诱导以MS培养基最好,不定芽的平均增值系数超过4;MS中加入6~BA3~4mg·L^-1+NAA0.1~0.5mg·L^-1时不定芽的诱导率接近60%,平均分化系数可达6.0以上;35g·L^-1的蔗糖浓度能有效的防止不定芽的玻璃化;垂直摆放有利于不定芽的分化;不定芽继代增值的最佳培养基为:MS+6-BA0.3mg·L^-1+NAA0.05mg·L^-1。 The mature embryo of Pinus bangeana was used as explants to induce adventitious buds and form plantlets. The thesis studied the use of mediums, hormone, concentration of sugar and effect of embryo inoculation in adventitious buds of Pinus bangeana proliferation and growth. The results showed that the best medium for adventitious bud inducement is MS, the adventitious buds proliferation rate was above 4. MS adding with 3 - 4 mg·L^-1 6- BA and 0. 1 - 0.5 mg·L^-1 NAA for a nearly 60% induction rate. The mean proliferation rate of adventitious bud reaches 6.0 . 35 g·L^-1 grams of sugar concentration can effectively prevent the glass. Vertical display of adventitious buds better differentiation. Following the value - added and the best medium was MS with 0.3 mg·L^-1 6-BA +0.05 mg·L^-1 NAA .
