The expression of PI3K/Akt signal pathway and HIF-1α in gastric carcinoma

摘要 目的:探讨PI3K、p-Akt及低氧诱导因子-1α(hypoxiainduciblefactor-1a,HIF-1α)在胃癌、癌旁和远癌组织中的表达。方法:收集胃癌切除术患者胃癌、癌旁及远癌组织标本,应用免疫组化法检测PI3K、p-Akt、HIF-1α蛋白的表达。结果:PI3K、p-Akt、HIF-1α蛋白在胃癌组织中表达最强(其灰度值分别为176.3±11.3,158.6±20.1,157.5±12.3),在远癌组表达最弱(196.9±7.4,192.3±8.3,194.2±5.3),在癌旁组表达介于上述两组之间(187.9±5.1,173.9±14.3,176.0±7.4),三组间差异有统计学意义(P<0.05)。HIF-1α的表达与PI3K、p-Akt呈正相关(r=0.662,0.674)(P<0.05)。结论:PI3K、p-Akt、HIF-1α在胃癌组织中高表达,且HIF-1α表达与PI3K、p-Akt表达呈正相关。 Objective : To explore the expressions of PI3 K, p - Akt and hypoxia inducible factor - 1 α ( HIF - 1 α) in gastric carcinoma, adjacent tussue and distal gastric tissue and their relation. Methods:Gastric carcinoma, adjacent tissue and distal gastric tissue sample were obtained from gastric carcinoma cases underwent gastrectomy. PI3K, p - Akt and HIF - 1 α protein expressions were examined by immunohistochemical method. Results:The expressions of PI3 K, p - Akt and HIF - 1 α protein were highest in gastric carcinoma group ( gray value 176.3 ±11.3,158.6 ± 20.1, 157.5 ± 12.3 ) ,were lowest in distal gastric tissue group ( 196.9 ± 7.4,192.3 ± 8.3,194.2 ± 5.3 ), and were intermediate in adjacent tissues ( 187.9 ± 5.1,173.9 ± 14.3,176.0 ± 7.4 ). The differences among these three groups were significant (P 〈 0. 05 ). The expression of HIF - 1 α were positively correlated with PI3 K and p - Akt expressions (r =0.662,0.674) (P 〈0.05). Conclusion: PI3K,p - Akt and HIF - 1α are overexpressed in gastric carcinoma. The expression of HIF- 1α is positive correlated with PI3K and p- Akt expressions in gastric carcinoma.
