摘要
目的:建立甘遂中大戟二烯醇及其同分异构体表大戟二烯醇的含量测定方法。方法:样品以乙酸乙酯为提取溶剂,超声处理后,蒸干,加甲醇溶解,以HPLC法分析。采用Agilent XDB C8(150mm×4.6mm,5μm)色谱柱分离,柱温30℃;以乙腈-水(95∶5)为流动相,流速1.0mL·min-1;检测波长210nm。结果:大戟二烯醇和表大戟二烯醇的线性范围分别为0.43~6.34μg(r=0.9999)和0.21~3.17μg(r=0.9999),平均加样回收率(n=9)分别为100.0%和102.2%。10批次甘遂中,大戟二烯醇和表大戟二烯醇的含量分别为0.18%~0.68%和0.12%~0.39%。结论:本方法准确、可靠,具有良好的重复性和稳定性,可用于甘遂中大戟二烯醇及其同分异构体表大戟二烯醇的定量分析。
Objective: To develop an HPLC method for simultaneous determination of euphol and tirucallol in Radix Kansui. Methods:The sample was extracted with ethyl acetate by uhrasonication. HPLC analysis was performed on an Agilent XDB C8 column (150 mmx 4. 6 mm ,5 μm). The mobile phase was the system of aeetonitrile -water(95 :5). The flow rate was 1.0 mL ·min-1 and UV detection was set at 210 nm. Results:Euphol and tirueallol had good linearity in the ranges of 0.43 - 6.34 μg ( r = 0. 9999 ) ,0.21 - 3.17 μg ( r = 0. 9999 ), respectively; The aver- age recoveries( n = 9 ) were 100.0% and 102.2% , respectively. The contents of euphol and tirueallol in Radix Kan- sui were O. 18% -0.68% and 0.12% -0.39% ,respectively. Conclusion:The developed method is accurate with high repeatability,which is helpful for the determination of euphol and tirucallol in Radix Kansui.
