摘要
目的:建立高效液相色谱法测定解表清肺丸中柚皮苷、橙皮苷、黄芩苷3种成分的含量。方法:采用ZORBAX SB-C18(4.6mm×250mm,5μm)色谱柱,以0.05%磷酸溶液(含2%乙腈)-乙腈为流动相进行梯度洗脱,流速1mL·min-1,检测波长277nm,柱温40℃,进样量10μL,外标法定量测定柚皮苷、橙皮苷、黄芩苷3个成分的含量。结果:柚皮苷、橙皮苷与黄芩苷浓度分别在5.26~52.5μg·mL-1(r=0.9999),5.12~30.7μg·mL-1(r=0.9999),10.0~60.2μg·mL-1(r=0.9998)范围内线性关系良好;平均回收率(n=6)分别为101.7%(RSD=0.54%),99.2%(RSD=1.3%),101.3%(RSD=1.0%)。结论:本法结果准确,灵敏度高,重复性好,方法简便可行,可用于测定解表清肺丸中柚皮苷、橙皮苷与黄芩苷3个成分的含量。
Objective: To develop an HPLC method for determination of three constituents ( naringin, hesperidin, ba- icalin)in Jiebiao Qingfei pills. Methods: Analysis was carried out on a ZORBAX SB C18 column (4. 6 mm ×250mm,5μm) eluted with water containing 0. 05% phosphoric acid and 2% acetonitrile -acetonitrile as the mobile phase in the gradient elution at the flow rate of 1 mL ·min-l. The UV detection wavelength was 277 nm, and the temperature of column was 40 ℃ The sample injection volume was 10μL. The contents of naringin, hesperidin and baicalin were calculated by external standard method. Results: The linear ranges of naringin, hesperidin and baicalin were 5.26 -52.5 μg · mL-1 (r =0. 9999) ,5.12 -30. 7 μg · mL-1 (r =0. 9999) ,and 10. 0 -60. 2 μg · mL-1 (r = 0. 9998 ), respectively;The average recoveries were 101.7 % (RSD = 0.54% ) ,99. 2% (RSD = 1.3 % ), 101.3 % (RSD = 1.0% ), respectively. Conclusion: The HPLC method is simple, reliable and specific with good reproduc- ibility, and can be used for determination of three constituents in Jiebiao Qingfei pills.
