抗CD_3单克隆抗体的制备、标记与鉴定

Production,Labeling and Identification of Anti CD 3 Monoclonal Antibodies

选用抗人Ｔ细胞ＣＤ３杂交瘤细胞产生的腹水，经系列免疫化学方法纯化和鉴定后，以异硫氰酸荧光素（ＦＩＴＣ）标记纯化之抗ＣＤ３单克隆抗体。将构建的荧光抗体与标准抗ＣＤ３单克隆抗体在流式细胞仪上分别对同一样本进行检测共２４例，ＣＤ３＋细胞分别是（６３８±４３）％和（６８２±４７）％，两者差异无显著性（Ｐ＞００５）。

Anti CD 3 McAb were purified from ascitic fluid of BACA/C mice induced by anti CD 3 hybridoma cell line. After identification,the anti CD 3 McAbs were labeled with FITC and used on flow cytometry for the detection of CD + 3 cells. The avevage detected CD + 3 cells from 24 blood samples was (63.8±4.3)%,compared with (68.2±4.7)% in the control group using standard anti CD 3 McAbs. No significant difference between two groups was found (P>0.05). This result suggested that the self constructed anti CD 3 McAbs was as good as standard one for the detection of CD + 3 cells on flow cytometry.
