快速DNA提取法在检测卡氏肺孢子虫PCR中的应用

STUDY ON THE RAPID METHOD OF EXTRACTED DNA FOR DETECTINING PNEUMOCYSTIS CARINII USING POLYMERASE CHAIN REACTION

为寻求一种适用于临床诊断的PCR方法。方法用快速PCR模板DNA制备方法提取实验大鼠肺组织，支气管肿泡灌洗液和血液中的卡氏肺孢子虫DNA，进行PCR扩增，并与传统的DNA提取法进行对比。结果两种方法收到一致的理想结果。结论快速法提取的DNA适用于卡氏肺孢子虫的PCR检测。将此法试用于临床病例检测，也收到如期结果，初步显示其推广应用前景。

Aim For simplifying the procedure of detecting Pneumocystis carniip (PC)using polymerasechain reaction (PCR). Method We extracted DNA by using a simple and rapid method from experimentalinfect-ed rat's lung tissue, bronchi--alveolar lavage (BAL), blood and patient's BAL. A comparison of the method withphenol--chloroform extraction ethanol precipitation method. Result The result demonstrted agreement betweenthe two method. Concluding The conclude that the simple and rapid method of extracted DNA is a valuable toolfor PCR detecting PC.
